Epithalon Before And After: Why Results Claims Resist Numbers

An epithalon before and after report has to clear two problems: an endpoint that moves too slowly to see, and a course whose mass was never recorded.

The most rigorous epithalon before and after account you will find looks like this: a ten day course, a telomere length test at each end, and the pair of numbers posted. It still fails to establish what it appears to establish, for reasons that have nothing to do with the poster's honesty.

Two separate problems have to be cleared. The endpoint has to be measurable at both ends by the same method, on a timescale where a change could plausibly appear. And the exposure has to be stated as a mass, which requires a number almost no account contains. This page takes both, in that order, and works the second one out in full. It gives no figure for a person.

The certificate for batch 15-05260628 assays this vial at 9.64 mg against a 10 mg label, inside the stated 10 percent tolerance, which puts the real figure at $2.59/mg on that batch. It carries purity, identity, endotoxin, sterility and heavy metals. Buying 3, 5 or 10 takes 3%, 5% or 10% off list.

An epithalon before and after report faces an endpoint problem first

Most before and after topics have an outcome you could photograph or weigh. This compound is sold against claims about ageing, which is the hardest possible category to observe over the length of a course.

Nothing about ageing changes measurably in ten days. So a report covering that interval is either reporting something else, such as sleep or mood, or reporting a laboratory value chosen because it moves faster than the thing it is supposed to represent. Both routes have problems, and the second one has the more interesting problem.

Telomere length as a before and after measurement

The obvious endpoint, given the claims associated with this compound, is telomere length. Treat it as a measurement and the difficulties appear immediately.

It is measured in a sample of cells, usually from blood, and the value depends on which cells were in the sample. A change between two draws can reflect a different mix of cell populations rather than a change in any cell. Assay methods differ, and a value from one laboratory is not directly comparable with a value from another. Repeat measurement on the same person with the same method produces a spread even with no intervention at all, so a difference between two single measurements is not automatically a change.

None of that makes the test useless. It makes a two point comparison, taken by one unblinded person who paid for both tests and expects a result, a weak instrument for detecting a small effect. A before and after pair is exactly that comparison.

There is also the question of what the endpoint would mean if it did move. Telomerase work associated with this compound is largely in cultured human cells, and cell culture is a mechanism observation rather than an outcome in a person. A shifted number in a blood sample is not a demonstration that anything about ageing changed.

Recomputing the mass behind a reported course

Set the endpoint aside and look at the exposure, which is the part arithmetic can fix.

A typical account says something like twenty units a day for ten days. That is a volume instruction, and it becomes a mass only when a concentration is supplied. Twenty units on a U-100 syringe is 0.2 mL, and that is all the account has actually specified.

At 5 mg per mL, 0.2 mL holds 1 mg, and ten days is 10 mg. At 10 mg per mL, the same 0.2 mL holds 2 mg, and ten days is 20 mg. At 25 mg per mL it holds 5 mg, and ten days is 50 mg. One sentence, three exposures, a factor of five between the ends.

Two reports, two fill volumes, one printed reading

Take two accounts written in identical words: twenty units daily, ten days.

The first bought a 10 mg vial and added 2 mL of bacteriostatic water. Concentration is 10 divided by 2, which is 5 mg per mL. Twenty units is 0.2 mL and 1 mg. Ten days is 10 mg, and the vial is empty, because 2 mL divided by 0.2 mL is exactly ten withdrawals.

The second bought a 50 mg vial and added 5 mL. Concentration is 50 divided by 5, which is 10 mg per mL. Twenty units is 0.2 mL and 2 mg. Ten days is 20 mg, and 3 mL of solution remains in the vial.

Two accounts, one printed reading, twice the mass in the second. Neither poster misdescribed what they did. The field that separates them is the fill volume, and neither thought to mention it because at the time it did not feel like part of the instruction.

What the account statesWhat it fixesWhat is still unknown
Twenty units dailyA volume of 0.2 mLThe mass in that volume
Ten daysA count of withdrawalsThe total mass over the course
The vial ran outA relationship between fill volume and useNothing about concentration on its own
A telomere test at each endAn endpoint of a kindThe assay, the laboratory, the repeatability
An improvementA subjective conclusionEverything measurable about it

Only the first row of the middle column is solid, and it is solid about the wrong quantity.

The interval is long and almost nothing in it is controlled

Accounts for this compound often stretch across months, because the claims concern slow processes and posters know it.

A longer interval buys exposure to everything else. Seasons change, sleep changes, training changes, other supplements come and go, and any of those moves the markers people choose as endpoints. There is no comparison arm, no blinding, and no pre registration of what would count as a result, so the analysis is chosen after the numbers are in.

Regression to the mean deserves its own sentence, because it is the mechanism most likely to produce an apparent improvement in exactly this design. People are more likely to start something after an unusually poor measurement, and a repeat measurement will tend to land closer to their average whether or not anything was done in between.

What a defensible write up would carry

Six preparation fields, which take one line: labelled mass, volume of water added, the concentration those two produce, the syringe scale, the volume drawn per session, and the resulting mass. Then the total mass over the course, which is the number the account is really about and the one that is never printed.

Then the endpoint, stated with its method, the same laboratory at both ends, and an acknowledgement of how much two measurements can differ with no intervention. Then a list of what else changed during the interval, written before the results were known rather than recalled afterwards.

Almost no account carries any of this. Supplying the six fields raises a post from meaningless to interpretable, and interpretable is still a single unblinded self report. It does not become evidence by being detailed, and it does not become evidence by agreeing with other posts, since the posts share their sources and their expectations.

That ceiling is worth naming, because the compound's own literature sits under a related limit. There are no registered interventional trials for it, so no trial identifier appears here, and it holds no marketing authorisation anywhere. The published work is largely associated with Khavinson and colleagues in St Petersburg, in cell culture, in mice and rats, and in human observational work from within that programme, with limited independent replication. Life extension is not demonstrated in people. A forum account cannot repair that, and a well written one does not come close.

Frequently Asked Questions

Can two accounts be compared if both state units?expand_more

Only if both also state a concentration. Units are a volume, and comparing volumes across different fill volumes compares nothing.

Would a telomere test at each end settle it for one person?expand_more

No. Two measurements from one unblinded person, possibly on different assays, cannot separate a real change from assay variability and sampling differences.

Does a longer account carry more weight?expand_more

It carries more exposure to confounding. Length is not control, and over months the number of uncontrolled variables grows faster than the evidence does.

If several accounts agree, is that worth something?expand_more

They agree about a wording, not about a mass, and they were written by people reading the same product pages. Repetition of an undefined exposure does not accumulate into a measurement.

What is the single most useful thing a poster could add?expand_more

The fill volume. It converts every other number in the account from a gesture into a quantity, and it costs one line to write down at the time.
