FOXO4-DRI Protocol: The Mouse Numbers, and Nothing Else
There is no clinical FOXO4-DRI protocol. What the published mouse experiments actually ran, why dosing was intermittent, and what a protocol would require.

A FOXO4-DRI protocol in the clinical sense does not exist. No human trial has been designed, approved or registered, so there is no eligibility criteria, no dose, no schedule and no monitoring plan for a person.
What does exist is the experimental design from the animal literature, and the arithmetic of turning a vial into a working solution. This page covers both, and gives no human schedule because there is none to give.
Where the assayed figures come from

The two published certificates cover different batches and disagree on net content: Kovera Labs assays batch 55-05260628 at 11.41 mg, MZ Biolabs assays lot 55-01260229 at 8.30 mg, both against a 10 mg label. Two different documents both called a protocol
The search traffic conflates two things, and separating them resolves most of the confusion.
A clinical protocol specifies eligibility, dose, route, schedule, endpoints, safety monitoring and stopping rules, is reviewed by an ethics committee, and is registered before enrolment. None exists for this compound anywhere.
An experimental protocol is the methods section of a laboratory study: model system, compound, dose, schedule, and how the outcome was measured. Several exist and are public.
Material circulating as a "FOXO4-DRI protocol PDF" is neither. It is generally compiled by a third party extrapolating from animal papers, presented with confidence the evidence does not support.
What the published experiments ran
The foundational 2017 study used several arms rather than one.

Two features are consistently misreported. The dosing was intermittent, three administrations across five days rather than a continuous course. And the routes used, intravenous and intraperitoneal, are laboratory routes in laboratory animals.
Why the schedule was short
The alternating-day pattern is not an arbitrary experimental choice, and reading it as a cycle gets the logic backwards.
A conventional drug is dosed continuously to hold a pathway suppressed; stop and the effect fades. A senolytic is not maintaining anything. It eliminates a cell population, and once those cells are cleared there is nothing further to act on until they re-accumulate, which happens over months and years.
That produces hit-and-run dosing: a short exposure sufficient to trigger apoptosis in the target population, then withdrawal. Continuous exposure would add risk without benefit, since healthy cells are present throughout and selectivity is relative rather than absolute.
The arithmetic a protocol would need, and what breaks it
Any experimental protocol starts from a known concentration, which is where this compound causes trouble.
Concentration is mass divided by volume. The mass most people use is the label, and for this product the label and the assays disagree: Kovera Labs measured 11.41 mg on batch 55-05260628, MZ Biolabs measured 8.30 mg on lot 55-01260229, both against 10 mg printed.

What would have to happen for a human protocol to exist
Preclinical toxicology across species establishing a safety margin. Then a phase 1 trial whose purpose is tolerability, dose-ranging and pharmacokinetics rather than efficacy: how long the peptide persists, how it is cleared, what dose produces unacceptable effects. Only then does anyone have a defensible starting dose for testing an outcome.
None of this has begun. A programme attempting it would also need to address a specific finding rather than merely proceed carefully: a 2023 study reported that clearing senescent cells could promote the development and progression of pulmonary hypertension, so the safety question is not purely theoretical.
