A NAD+ before and after account usually contains one number and three silences. Two posts appear in the same thread within a week of each other. The first says fifty units from a 500 mg vial and reports feeling sharper by the afternoon. The second says fifty units from a 500 mg vial and reports nothing at all. Read as written, these are two people who did the same thing and got different results, which is how the thread will treat them.

They almost certainly did not do the same thing. The sentence they both wrote is compatible with masses that differ by a factor of four, and neither post contains the field that would separate them.

Why a NAD+ before and after post cannot be converted to a mass

The missing field is the fill volume. A unit is a volume, not a mass: on a U-100 syringe it is 0.01 mL, and it stays 0.01 mL whatever is dissolved in it. Turning fifty units into milligrams requires a concentration, and the concentration was decided by whoever added the water, in private, before the post was written.

Four rows, one sentence, and a range from 25 mg to 100 mg. Note the last two rows in particular: different labels, different fill volumes, identical drawn mass. The vial size in the post correlates with nothing.

This is not a subtlety specific to careless posters. It is structural. The only number a syringe can report is a volume, and volume becomes mass only through a ratio that lives in a memory or on a scrap of paper. A post that quotes units is quoting the one field that survives the process, and it is the field that means least on its own.

Four vials, one identical sentence

Push the same arithmetic the other way and the failure becomes sharper. Suppose the two posters wanted to compare properly and agreed to state their masses instead of their unit readings. Both write 50 mg.

That still does not fix it, because a mass drawn from an unassayed vial is a mass claimed by a label. The 500 mg printed on the glass is a seller's statement, not a measurement, and injected NAD+ has no marketing authorisation from any regulator to sit behind it. Both posters computed honestly from a number neither could check.

There is a second distortion underneath. At these masses the powder occupies enough volume to matter. Take an assumed solid density of 1.5 g per mL, stated as an assumed input so the arithmetic can run: 500 mg then displaces roughly 0.33 mL. A poster who added 5 mL of water and computed 100 mg per mL was working with something nearer 94 mg per mL, so the 50 mg they reported was closer to 47. On a 5 mg peptide vial that correction is invisible. Here it is real, it points the same way every time, and nobody applies it.

The word in the post may not name the intervention

Now the problem that belongs to this compound and to almost nothing else on this site. A before and after account tagged with the letters NAD may be describing any of several different things.

These are not variations on one intervention. Oral precursors are swallowed and converted internally; injected material is not. The human interventional evidence sits mainly with the oral precursors, where nicotinamide riboside has been shown in human trials to raise blood NAD levels. The injected form has a much thinner record. A thread that pools accounts across those categories is averaging over a difference larger than any difference it is trying to detect.

So a reader collecting before and after reports is not building a sample. They are building a mixture whose composition they cannot recover, and the mixing happened before the first post was written.

The after is a feeling reported on the same day

Even a perfectly documented account runs into the outcome. The reported endpoints in this area are typically energy, clarity, mood and sleep, assessed by the person who chose to buy the product, often within hours of the first use, and with no blinded comparison anywhere in the process.

Those endpoints move on their own. They move with sleep the night before, with caffeine, with the weather, and with having just done something deliberate about one's health, which is not a small effect and has never been separated out in any account of this kind. A same day self report against an unblinded expectation is the measurement design most vulnerable to all of that, and it is the design every one of these posts uses.

There is also nothing external to check against. For a compound the body already makes and regulates, an account has no independent marker to point at. The person cannot show a level, and if they could, a level is a biomarker rather than an outcome.

What would make an account arithmetically usable

A short list, and it is short because the bar is low and still almost never met.

The vial label and the exact fill volume, because those two produce the concentration. The syringe scale, because a unit has no volume until the barrel is named. The volume drawn, in units or millilitres. The intervention, named as injected material or as a specific oral precursor, since the two are not comparable. The interval and the number of draws before the report. And the date the vial was reconstituted, because a solution drawn on day forty is not the same object as one drawn on day two.

Six fields, none of them hard to write down, all of them derivable at the bench. An account carrying all six could be compared with another account carrying all six. It would still not establish that the compound did anything, because a comparison of two unblinded self reports is not a trial. It would at least be a comparison of the same quantity.

Two posts agreeing on a number they both computed differently

The last trap is agreement. When two accounts both say 50 mg and both report improvement, the coincidence reads as corroboration.

Work backwards and the agreement dissolves. One poster reached 50 mg from a 500 mg vial in 5 mL, the other from a 1000 mg vial in 10 mL, and a third could reach it from 1000 mg in 5 mL at half the unit reading. Same figure, three routes, three different displacement errors, three different label claims, three different solution ages. The number they share is the output of division, and division agreeing does not make the inputs agree.

Two accounts that match tell you the arithmetic was done the same way. They tell you nothing about the material, and nothing at all about the outcome.
